Milk fat globule membrane – a source of polar lipids for colon health? A review

Abstract

The milk fat globule membrane (MFGM) surrounds fat globules, protects them against lipolysis and disperses the milk fat in the milk plasma. Besides their structural and emulsifying roles, in vivo and in vitro studies have demonstrated that phospholipids and sphingolipids of MFGM possess cancer risk-reducing properties. Several reports attribute its chemopreventive activity to products of sphingomyelin hydrolysis, which affect multiple cellular targets that control cell growth, differentiation and apoptosis. With knowledge on the potential health benefits of MFGM lipids and proteins, dairy industries could in the future address their research in developing new functional dairy products enriched in beneficial MFGM components.

Targeting sphingosine-1-phosphate signaling in lung diseases.

Abstract

Sphingosine-1-phosphate (S1P), a simple, bioactive sphingolipid metabolite, plays a key role, both intracellularly and extracellularly, in various cellular processes such as proliferation, survival, migration, inflammation, angiogenesis, and endothelial barrier integrity. The cellular S1P level is low and is tightly regulated by its synthesis and degradation.
 Sphingosine Kinases (SphKs) 1 and 2, catalyze the ATP-dependent phosphorylation of So, (sphingosine) to S1P, while the degradation is mediated by the reversible dephosphorylation catalyzed by the S1P phosphatases and lipid phosphate phosphatases and the irreversible degradation to hexadecenal (aldehydi)  and ethanolamine phosphate (EAP)  by sphingosine-1-phosphate lyase (S1PL).

 As a ligand for specific G-protein-coupled receptors, S1P1-5, which are differentially expressed in different cell types, S1P generates downstream signals that play crucial role in developmental and disease related pathologies. In addition to acting extracellularly on receptors located on the plasma membrane, S1P can also act intracellularly, independently of S1P1-5, affecting calcium homeostasis and cell proliferation.

 The SphKs /S1P /S1PL metabolic pathway is implicated in numerous human pathologies including respiratory disorders, thereby raising the possibility that manipulating intracellular S1P levels could offer therapeutic potential in ameliorating lung diseases. This review focuses on the prospects of targeting S1P signaling and S1P metabolizing enzymes using small molecule inhibitors, receptor agonists, and antagonists in the treatment of lung diseases.

TIIVISTELMÄ, Abstract

 Lyhytketjuiset tyydyttyneet rasvahapot etikkahappo C2:0), propionihappo(C3:0) ja voihappo( C4:0)  ovat bakteerien aineenvaihduntatuotteita, jotka välittävät vuorovaikutusta dieetin, mikrobifloran  ja isäntäkehon kesken. 


  • The short-chain fatty acids (SCFAs), acetate, propionate and butyrate are bacterial metabolites that mediate the interaction between diet, the microbiota and the host. 
 Tässä artikkelissa  mitattiin määrällisesti  SCFA- rasvahappojen  systeeminen saatavuus ja niiden  sijoittuminen biologisesti relevantteihin molekyyleihin.
  • In this study, the systemic availability of SCFAs and their incorporation into biologically relevant molecules was quantified.
 Tutkimuksessa merkattiin  13C-isotoopeilla(*) etikkahappoa, propionihappoa ja voihappoa ja niitä asetettiin colonalueelle 12 terveelle henkilölle kapseleissa ja mitattiin   isotoopilla (*) merkattujen tyydyttyneiden rasvahappojen (*SCFA)  ja *glukoosin,  *kolesterolin  ja rasvahappojen (*FA)  plasmapitoisuudet. Samoin mitattiin  suolistofloran kyky tuottaa voihappoa (C4:0).
  • Known amounts of 13 C-labelled acetate, propionate and butyrate were introduced in the colon of 12 healthy subjects using colon delivery capsules and plasma levels of 13 C-SCFAs and of 13 C-glucose, 13 C-cholesterol and 13 C-fatty acids were measured. The butyrate producing capacity of the intestinal microbiota was quantified as well. 
 Colonalueelle asetettujen *rasvahappojen  systeeminen  saatavuus oli *etikkahapolla 36 %, *propionihapolla 9% ja *voihapolla 2%.Se interkonversio , mikä näissä hapoissa oli tavallisinta, oli *etikkahapon muuttuminen *voihapoksi (24%) colon- mikrobifloran vaikutuksesta eikä se korreloinut fekaalinäytteiden voihapontuottokapasiteettiin.
  • Systemic availability of colonic-administered acetate, propionate and butyrate was 36%, 9% and 2%, respectively. Conversion of acetate into butyrate (24%) was the most prevalent interconversion by the colonic microbiota and was not related to the butyrate-producing capacity in the faecal samples. 
Alle 1%  merkatusta *etikkahaposta asettui *kolesteroliin ja alle 15% asettui *rasvahappoihin.  Coloniin  asetetusta  *propionihaposta asettui *glukoosiin 6%.  SCFA-rasvahapot* erittyivät  pääasiassa keuhkojen kautta isotooppia kantavaksi  CO2- hiilidioksidiksi* oksidoitumisen  jälkeen, kun taas  alle 0,05%  SCFA*:sta  erittyi virtsaan.
  • Less than 1% of administered acetate was incorporated into cholesterol and < 15% in fatty acids. On average, 6% of colonic propionate was incorporated into glucose. The SCFAs were mainly excreted via the lungs after oxidation to 13 CO2 whereas less than 0.05% of the SCFAs were excreted into urine. 
 Nämä tulokset   tekevät mahdolliseksi arvioida  ja mitata määrällisesti  tulevaisuudessa SCFA- rasvahappojen tuotantoa ihmisen colonissa   C13-isotoopilla merkatuista kuiduista* mittaamalla  merkattujen SCFA*- rasvahappojen pitoisuudet verestä.
  • These results will allow future evaluation and quantification of SCFAs production from 13 C-labelled fibres in the human colon by measuring 13 C-labelled SCFA concentrations in blood.

Abstract

Inorganic polyphosphate (polyP) was previously identified as a probiotic-derived substance that enhances intestinal barrier function. PolyP-accumulating bacteria are expected to have beneficial effects on the human gastrointestinal tract. In this study, we selected Lactobacillus paracasei JCM 1163 as a strain with the potential to accumulate polyP, because among the probiotic bacteria stored in our laboratory, it had the largest amount of polyP. The chain length of polyP accumulated in L. paracasei JCM 1163 was approximately 700 phosphate (Pi) residues. L. paracasei JCM 1163 accumulated polyP when Pi was added to Pi-starved cells. We further improved the ability of L. paracasei JCM 1163 to accumulate polyP by nitrosoguanidine mutagenesis. The mutant accumulated polyP at a level of 1500 nmol/mg protein-approximately 190 times that of the wild-type strain. PolyP extracted from the L. paracasei JCM 1163 significantly suppressed the oxidant-induced intestinal permeability in mouse small intestine. In conclusion, we have succeeded in breeding the polyP-accumulating Lactobacillus mutant that is expected to enhance intestinal barrier function.

 On käytetty aktivoituja lieteprosesseja anaerobista ja aerobista tilaa vaihdellen ( anaerobinen-aerobinen prosessi)  ja on saatu menestyksellä  tehostettua  biologisen fosfaatin poistamista    jätevedestä. (EBPR)
  • Activated sludge (liete)  processes with alternating anaerobic and aerobic conditions (the anaerobic-aerobic process) have been successfully used for enhanced biological phosphate removal (EBPR) from wastewater. 
 On tiedetty, että polyfosfaattia (PolyP)  akkumuloivat bakteerit (PAB) ovat  välttämättömoä EBPR:ssa anaerobisaerobisessa prosessissa.
  • It is known that polyphosphate-accumulating bacteria (PAB) play an essential role for EBPR in the anaerobic-aerobic process.

  •  Exposure of microorganisms to alternate carbon-rich anaerobic environments and carbon-poor aerobic environments in the anaerobic-aerobic process induces the key metabolic characteristics of PAB, which include organic substrate uptake followed by its conversion to stored polyhydroxyalkanoate (PHA) and hydrolysis of intracellular polyphosphate accompanied by subsequent Pi release under anaerobic conditions.
Intrasellulaarisen glykogeenin oletetaan olevan  solun redoxtasapainon säätelijä. Glykogeenin varastoiminen  on PAB- organismien avainstrategiaa, jolla  ne pitävät redoxtasapainonsa  ottaessaan anaerobisesti erilaisia orgaanisia aineksia sisäänsä  ja täten ne voittavat  mikrobiologisessa valinnassa.
  • Intracellular glycogen is assumed to function as a regulator of the redox balance in the cell. Storage of glycogen is a key strategy for PAB to maintain the redox balance in the anaerobic uptake of various organic substrates, and hence to win in the microbial selection. 
 Akinetobakteerilajit, Mikrolunatus fosfoforus, Lampropedia lajit ja Rhodosyklus-ryhmä  ovat raportoituja PAB-ehdokkaita.  Saattaa olla niin, että PAB ei käsitä  harvoja rajallisia geneettisiä lajeja, vaan niisä on  fylogeneettisesti ja taksonomisesti erilaisia bakteeriryhmiä.
  • Acinetobacter spp., Microlunatus phosphovorus, Lampropedia spp., and the Rhodocyclus group have been reported as candidates of PAB. PAB may not be composed of a few limited genospecies, but involve phylogenetically and taxonomically diverse groups of bacteria. 
Jotta voitaisiin määritellä EBPR-prosessien  mikrobiyhteiskunnan rakenne, pitäisi tarkastella lähemmin jokaisen PAB-lajin ilmenemistä ja käyttäytymistä erilaisissa EBPR-prosesseissa pääasiassa molekulaarisin menetelmin, koska useaa  PAB -lajia näyttää olevan mahdotonta viljellä.
  • To define microbial community structure of EBPR processes, it is needed to look more closely into the occurrence and behavior of each species of PAB in various EBPR processes mainly by molecular methods because many of PAB seem to be impossible to culture.

 Of theoretical interest regarding poly P is its antiquity in prebiotic evolution, which along with its high energy and phosphate content, make it a plausible precursor to RNA, DNA, and proteins. Practical interest in poly P includes many industrial applications, among which is the microbial removal of Pi in aquatic environments.

Malonaattijoni  ( propanedioate)  on hahmolta meripihkahapon tapainen dikarboksyylihappo, mutta siltä puuttuu   CH2-CH2- ryhmä, joka voisi osallistua dehydrogenaatioreaktioon kuten meripihkhappo voi (succinate). Wikipediasta löytyy siihenkin valmis ajatus kirjallisessa muodossa.Malonyl CO ei ole ensin "malonihappo joka aktivoituu"  vaan se syntetisoituu  aktivoidusta etikkahaposta   ja biotiinin avustuksella lisätään CO2- hiilidioksidiryhmä ( bikarbonaatista) , jolloin muodostuu malonyyli-CoA. Se on tarkasti säädelty  aineenvaihdunnallinen moduli, lego ja MCAT-entsyymi  siirtää tästä malonaatti-osan  (Asyyli-ntsyymii-kompleksiin tai asetyyli-entsyymiin-komplekiin ja niin saadaan  3C pidempi molekyyli ensin, muta  se kondensoituu ja irrotaayhden CO2- ryhmän., joka kuului malonihappoon ja oli juuri se joka oli lisätty biotiinilla. 
Tämä lähde kertoo, että MCAT entsyymi  osallistuu  myös polyketidisynteesiin , ketomuodoista polymerisoituja . molekyylejä luonnossa kuten bakteereissa. tässä artikkeli maintise niille entsyymin PKS, polyketidisyntetaasin.   mutta ihmisestä tiedetään nyt ainakin  että  4C kokoa   beta-OH-muoto voihappovaiheesta - voi  polymerisoitua . (PHB). Spontaanistiko?  Fosfaatithan osallsituvat  puskuritoimintaan soluissa ja polymeroituvat ketjut  ovat inerttejä valmiina.

Malonyl-CoA is a coenzyme A derivative of malonic acid.

1.1. It plays a key role in chain elongation in fatty acid biosynthesis and polyketide biosynthesis.
Malonyl-CoA is also used in transporting alpha-ketoglutarate across the mitochondrial membrane into the mitochondrial matrix.

Malonyl-CoA is formed by carboxylating acetyl-CoA using the enzyme acetyl-CoA carboxylase. One molecule of acetyl-CoA joins with a molecule of bicarbonate, requiring energy rendered from ATP.
Malonyl-CoA is utilised in fatty acid biosynthesis by the enzyme malonyl coenzyme A:acyl carrier protein transacylase (MCAT).

Polyketide biosynthesis

MCAT is also involved in bacterial polyketide biosynthesis. The enzyme MCAT together with an acyl carrier protein (ACP), and a polyketide synthase (PKS) and chain-length factor heterodimer, constitutes the minimal PKS of type II polyketides.

(Pohdinta   Samalla kun  malonaatti vähentää  energia-aineenvaihduntaa, se lisää  mahdollisuutta neutraalien rasvahappojen synteesiin , lisää palmitiiniallasta ja  vaikuttaa hiljentävästi beta-oksidaatioon-   ja täten  epäsuorasti  edistää glukoneogeneesia ja glykogeenin akkumulaatiota. Myös krebsin syklin alkupää on avoinna käytölle.

Malonyl-CoA is a highly-regulated molecule in fatty acid synthesis; as such, it inhibits the rate-limiting step in beta-oxidation of fatty acids.
 Malonyl CoA inhibits fatty acids from associating with carnitine by regulating the enzyme carnitine acyltransferase, thereby preventing them from entering the mitochondria, where fatty acid oxidation and degradation occur.

Kuten tekstistä ylhäältä huomaa, malonihappoa ei sinänsä mitenkään syödä niihin kohtiin, missä sitä kehossa tqarvitaan, vaan siä muodsotuu  kun on hyvä energiatasapino ja liikunta ja hapensaanti.  Se on itseasiasa moduli enemmän kuin molekyyli. Runsaat ylimääräiset hiiliketjut hiilihydraattien aineenvaihdunnasta  saadaan muokattua malonaattimuotoon, jolloin se on predestinoitunut tuotatmaan PALMITIINIHAPPOA tiettyyn  pisteeseen asti, kunnes palmitiinihappoa alkaa olla niin tarpeeksi että se alkaa ajrrutaa malonimodulin  synteesifunktiota.

Palmitiinihappo on erittäin tärkeä kehossa. varsinkin keuhkoille sen merkitys on suuri. Keuhko valmistaa spesifisesti paikalliset surfaktanttilesitiininsä  kahdesta palmitiinihaposta- dipalmityl-lesitiinejä.   Epäilen vähän kannattaako maitorasvoja jättää liian niukalle ravinnossa, sillä niistä tulee tukea kehon palmitiinihapposynteesiin niin että on hyvä immunologinen luonnollinen puolustukyky esim keuhkotulehduksia vastaan. Aina kun keuhkoissa on jotakin häikkää, kannattaa muistaa  lääkkeen tapaan tavallinen  täysmaito . vanha keino on keitettyä sipulimaitoa. Sipulista saa myös orgaanista rikkiä palmitiinien lisäksi, molemmat avustavat keuhkopuhdistumaa. .
Luonnollisesti keho tuottaa geneetttisesti  palmitiinia hiiliketjuista käsin riittävästi, . kun  ravinto on tasapainoista  ja riittävää ja kunnon ulkoilulla käytetän keuhkoja, että ne antavat signaalin keuhkolesitiinin tarpeesta ja bikarbonaattiakin on  saatavissa.  malonaatin synteesiin.
